There are no kids of Dean which we know of but i personally think that Dean is the father of Ben, the little boy from the episode "The Kids are Alright" but it was denied in that episode so no.

No, but in Season One I think it is, He thought he had a little boy named Ben par one of his past "conquests" who was very good bending & moving ;) LoL
Dean does have a daughter! I don't recall which episode this took place in, but I have seen this before. Dean had a daughter, however, Sam kills her. Look up "Dean has a daughter and Sam kills her" on YouTube, toi can find the scene there. He does not deny the girl who claims to be his child, he does acknowledge her as his child.

Ben isn't biologically Dean's - Lisa a dit that she'd had a test done and it came back positive for some other guy. She wouldn't have kept lying about that when Dean was living with them for an entire year!
