I agree with Castiel4ever though. and even though Sam gave it too him, I think Dean was just too depressed about everything and Lost his hope and faith (especially seeing what Sam's version of *his* heaven was...) a truly sad episode! I hope Sam takes it out of the trash and gives it back to him later!
I think he did it because he Lost faith. Like Joshua said, it was very important for Dean to find God. He hoped - he will help them but mostly to himself........because of that Sam's demon blood thing,how broken he are and Michael's thing...All that presure ..... and then sudenly they tell him to back of God will not help you. It's not his problem!! If I were Dean, I would do the same!!! That necklase was all his hope, solution to all his problem!!! disappointment was too big to keep it!!!

Yep i agree :) I think it was about loosing all hope.
In my view Dean threw away the amulet because of Sam. Sam had donné it to him and he has always cherished it not knowing anything about the god thing. But after going to heaven with Sam and seeing that Sam's heaven was completely about the times he spent away from Dean while Dean's were about great moments he had with Sam and the family he was hurt. I think that he did it knowing it would hurt Sam.
Dean's relationship with Sam at the time was poor. He had felt betrayed par Sam and thought they could never be what they have been to each other before..so his connection to it was not the same.
